Read-only is not necessary. It's just a failsafe so Steam or the game doesn't overwrite it with the default values.
Search for masterservers.vdf in your game directory and delete all of them. Sometimes there are duplicates of the masterservers.vdf file. Then follow the install instructions again.

You are doing something wrong. There are lots of other threads on these forums where people have had similar problems but they've mostly got it working by following the instructions. Search the other threads and you might get ideas why it isn't working for you.
Basically the masterservers.vdf is just a configuration file. If it's not there then hard coded default masterservers are used. Sometimes there might be duplicates of masterservers.vdf created by Steam or the game and those you need to delete. And then follow the Setti masterserver install instructions.

...SETTi really should add ("new") rev emu compatibility to the "Masterserver" how-to since there the masterserver.vdf is just mandatory, the emu itself contains the IP already
rev.ini -> #MasterServer=213.239.209.221 (its comented since the default value (old IP) is hardcoded in the emu)
rev.ini just needs to be edited to MasterServer=188.40.40.201 (NO # infront!) and voila. im not sure if rev finaly overtaken the new master IP into their latest emu, since they dont seem pretty afforted after they lost the source of *nix emu, but above applies at least upto v9.78 client emu.
else its just a 50:50 case the servers are getting queried from SETTi (Masterservers.vdf).
